    Hello Drop-seq community!

    The first question is when primers with linear amplification will be available?
    T7 proved to be superior to STRT, it is known since CEL-seq paper http://www.sciencedirect.com/science/article/pii/S2211124712002288
    Followed by MARS-seq.
    Of course switch at 5′ may look more user friendly, but the quality of the data is the key for analysis of low expressed genes, like transcription factors:
    The actual target of ours in the quest for neuronal diversity.

    How do we convince Chemgenes to synthesize beads with T7 promoter at 5′ of the illumina_adaptor_barcode-UMI- ???

    Hi Pavel,

    Thanks for your post. It would be interesting to try different amplification strategies with Drop-seq. ChemGenes will make barcoded beads with custom primer sequences. If you do order ones with a T7-compatible sequence and try them, let us know how your libraries look!
